Growth factors for dermal wounds

  • TGF-α (member of EGF family)
    • Potent keratinocyte mitogen and motogen
    • Released at site of injury
    • Not enough experimental data yet
  • FGF
    • Causes proliferation of keratinocytes and fibroblasts
    • Animal studies: FGF-2 caused reepithelialisation of wounds and enhanced healing of chronic pressure sores
    • Large pharmaceutical trials inconclusive
  • PDGF (AB-platelets and BB-macrophages and epidermal cells in response to injury)
    • Application to wounds led to more rapid healing
    • Clinical trials for diabetic foot ulcers encouraging
    • PDGF BB was first growth factor to be approved by FDA
  • TGF-β family:
    • Chemotactic for macrophages
    • Stimulates production of collagen 1 and fibronectin
    • 1 and 2 stimulates wound healing in animals
    • Human trials - TGFβ2 useful for venous stasis ulcers
